Development of high fluorescence, target-specific organic dye system is crucial for biological detection and imaging fields. To overcome the limited applications of organic dyes, dye-graft-polymer system was developed. Reactive polymer, poly(pentafluorophenyl acrylate), was utilized as backbone to immobilize near-infrared region dye, IR775. Hydrophilicity of the polymer was adjusted by additional substitution with amino-2-propanol or polyethylene glycol. Biotinylation of the polymer added target-specific binding ability to dye-graft-polymer system to bind with streptavidin protein. Absorbance and fluorescence spectra were examined for synthesized dye-graft-polymers and the grafting density of IR775 was optimized to 3%. Then, selectivity of biotinylated dye-graft-polymer toward streptavidin coated materials was examined. Biotinylated dye-graft-polymer selectively bound to streptavidin with high fluorescence signal. Biotinylated dye-graft-polymer system is expected to be efficient fluorescence system with high fluorescence, target-specific binding ability, and adjustable characteristics.
