Gene expression can be regulated by chromatin remodelers through nucleosome disassembly or rearrangement in promoter proximal regions or gene bodies. However, its mechanism is not well known. Here, I focused on Fft3 chromatin remodeler in Schizosaccharomyces pombe. By using Precision Nuclear Run-on sequencing (PRO-seq) that can detect active polymerase II with high resolution, I demonstrated that Fft3 was crucial in maintaining stable promoter proximal pausing, and without Fft3, premature transcription termination seemed to occur.
