High-throughput screening of amino acid production in Escherichia coli using its toxic analog유독성 유사체를 이용한 아미노산 생산 균주의 대량 탐색

Climate change is an important issue to be solved, so that various approaches have been performed to decrease and minimize the factor related to induce climate change. Especially, metabolic engineering has been paid enormous attentions due to its potential to replace petroleum industries and its eco-friendly development. However, one of bottlenecks in the development of metabolic engineering is a long time scale required to verify the engineering steps and therefore, achieve significant improvement in target production. To decrease the time and efforts required to examine the effect of genes on production of the target chemicals, high-throughput genomic regulation system has been developed. In this paper, we utilized one of high-throughput genomic regulation system tool, synthetic small regulatory RNA, to screen a library of approximately 1,800 genes at one time in the plate containing toxic analog of amino acid. Phenylalanine and proline producing strains were exemplified and productions of mutant resistance to toxic analog were increased.
