Understanding brain disorders using three-dimensional cellular assembly called organoid is actively researched because of organoid’s functional advantages over two-dimensional cell culture model and animal model. Because of organoid’s structural uniqueness, specialized sensor platform was required. We present a miniaturized transparent dopamine sensor platform able to monitor dopamine secretion from mid-brain organoid and imaging through transparent electrode. The miniaturization of 3-electrode system of the sensor was achieved using the micro-electromechanical system (MEMS) technology and biocompatible polydimethylsiloxane (PDMS) chambers with an equivalent size of the commercial well plate. The transparency was achieved using nanosphere lithography on working electrode. The dopamine released from mid-brain organoid using sensor platform was detected with different concentrations. This novel, simple, inexpensive and time-saying transparent dopamine sensor platform allows for detection of dopamine and imaging simultaneously.
