폐동맥고혈압 동물모델 및 이를 이용한 폐동맥고혈압 예방 또는 치료용 물질의 스크리닝 방법Animal model of pulmonary arterial hypertension and method for screening material for preventing or treating pulmonary arterial hypertension using the same
본 발명은 폐동맥고혈압 동물모델 및 이를 이용한 폐동맥고혈압 예방 또는 치료용 물질의 스크리닝 방법에 관한 것으로, 본 발명자들은 본 발명의 폐동맥고혈압 동물모델을 이용하여 Sox17 유전자의 기능 소실과 저산소증으로 폐혈관의 병적인 리모델링(pathologic vascular remodeling)과 우심실 리모델링(right cardiac remodeling)이 동반된 폐동맥고혈압을 효과적으로 유도할 수 있음을 확인하였고, 또한 저농도 산소에 노출된 폐동맥고혈압 동물모델에서 HGF(hepatocyte growth factor) 및 c-Met(tyrosine-protein kinase Met)의 발현 및/또는 활성이 증가하는 것을 확인하고, HGF 및 c-Met의 발현 또는 활성을 억제하는 경우 폐동맥고혈압 병증을 완화할 수 있음을 규명하였는바, 본 발명의 스크리닝 방법을 통해 폐동맥고혈압의 예방 또는 치료 물질을 신속 및 정확하게 발굴할 수 있고, 폐동맥고혈압 치료제를 효과적으로 개발 및 선별하는데 유용하게 이용할 수 있다.
