Developing the next-generation drug delivery system with gold nanorod금 나노로드 기반 차세대 약물 전달체 개발

The current precision personalized medicine approaches are focused on improving the precision to reduce the toxicity whether than achieving personalization at the individual level. Therefore, there is still a strong need for next-generation precision personalized medicine to control drug release based on the patient’s response. Thus, a novel drug delivery system (DDS) that could adjust the drug release rate from external light stimuli was developed by creating nanoparticles from gold nanorods (AuNR) with different aspect ratios of 1.56, 2.66, and 7.64. and absorbance peak at 650 nm, 800 nm, and 1100 nm, respectively. Moreover, helical AuNR was made to induce chirality in the DDS. The self-assembly of AuNR and helical AuNR into nanoparticles were caused by conjugating amphiphilic polymer, Thiol-poly (ethylene glycol)-b-poly(ε-caprolactone) to the surface and by using the water-in-oil emulsion method. Finally, through the successful emulation of AuNR to the microtubules in HL-60 cells, the feasibility of using a novel bio-templating approach known as the Conversion of Advanced Materials via Biostructure (CAMBIO) was confirmed. With CAMBIO advantage of cellular compartmentalization can be used to achieve efficient combination therapy delivery. These results set the ground for creating the next-generation personalized precision medicine DDS that can release drugs based on the patient’s response even after the drug is administered.
