Generation of uniform photo-crosslinkable hydrogels in microfluidic channels미세연성유체소자 내에서 균일한 광가교성 하이드로젤 제조

Monodisperse physical and chemical crosslinked micro-hydrogels have received much attention recently due to their potential capacities as drug carriers. Photo-crosslinkable amphiphilic block copolymers composed of poly(ethylene oxide)-b-poly(propylene oxide)-b-poly(ethylene oxide) as the backbone were synthesized by conjugating with acryloyl chloride substituting for two terminal hydroxyl groups. The synthesis was verified NMR and FT-IR analysis and the degree of substitution of acrylated groups at terminal site of Pluronic F127 copolymers was determined from $^1H-NMR$. For fabrication of uniform micro-hydrogels, a soft-microfluidic deivice was exploited by emulsifying an aqueous diacrylated Pluronic F127 phase in an olive oil phase containing ABIL EM 90 as a surfactant. The size of water_in_oil emulsion droplets were controlled by aqueous phase and oil phase flow rates, and surfactant concentration dissolved in olive oil phase. Droplet size increased as interface tension and flow rate of aqueous phase increased, or the flow rate of oil phase decreased. Emulsion droplets were also generated at regular frequency by the break-up process at the hydrodynamical focusing regime. The synthetic copolymers within pre-polymer emulsion droplets with narrow size distributions were thermally gelated flowing through a microfluidic channel, and chemically crosslinked by UV-exposure. They exhibited a 65~70 % volume shrinkage varying from at 4℃ to at 40℃, and showed a reversible swelling and deswelling behavior due to the hydrophobic interaction of poly(propylene oxide) middle blocks in the diacrylated Pluronic copolymers with a change of temperature. The release performance was verified by the change of intensity profiles of FITC-dextran loaded in the micro-hydrogel inferred from fluorescence spectra at different temperature (30℃, 40℃), resulting in exhibiting controlled release and leveling off after initial burst.
