YAP (Yes-associated protein) is regulated negatively through the phosphorylation of YAP S127 by Hippo pathway and Akt pathway. These negative regulations occur at each specific circumstance. Hippo pathway inhibits YAP when differentiation signal or contact inhibition induced, and Akt inhibits YAP when cells get survival signal. However, whether YAP at cycling cells-like cancer cell line is regulated or not, is not known. Here, I show that the phosphorylation profile of YAP S127 which controls its local distribution and activity is changed through cell cycle progression. Especially, the level of phosphor-S127 YAP is decreased in G1/S phase and gradually increased as cells progressed to G2-M phase after thymidine release in HeLa cells. On the contrary, nuclear YAP is increased in G1/S phase. And it was generalized through Nocodazole synchronization system in HeLa cells and serum starvation system in NIH 3T3 cells. I present whether the various contexts of YAP can induce the changes of normal cell cycle profiling and progression. So I transfected mutants of YAP and investigated the cell cycle profiling. To understand in more detail, I synchronized YAP mutant expressing cells to G1/S phase by thymidine block and investigated the cell cycle progression.
