dc.description.abstractCHAPTER 1. Characterization of Compact Protein Monolayers Constructed on Dendrimer Monolayers with Atomic Force Microscopy. Atomic Force microscopy를 이용한 덴드리머 고분자 단분자막 위에 형성된 밀집한 단백질 단분자막 분석 Ever since its invention, atomic force microscopy (AFM) has been widely used in biotechnology and biomedical research, including imaging, forcemapping, and sensing applications. In the previous works, our laboratory tried to construct effective protein microarray chip platform with PAMAM dendrimer, and in these process, we could observe the closely packed streptavidin layer on the dendrimer layer. Also, we could know this surface coverage, 88%, with Surface Plasmon Resonance (SPR) spectroscopy. So we examined the surface morphology of dendrimer/biotin and dendrimer/biotinlstreptavidin by Atomic Force Microscopy (AFM), and as a result, we could observe definite differences between two surfaces. Also we could observe the closely packed streptavidin layer image, and through the line profiling of captured image, we could confirm the streptavidin single molecule image, indirectly. CHAPTER 2. Development of the Biospecific Chip Platform on a Glass using Poly(amidoamine) Dendrimer. 덴드리머 고분자를 이용한 유리단백질 칩의 개발 As the human genome has been thoroughly discovered, complementary understanding of protein function and interaction, proteomics, is essential and necessary. For these researches, there were lots of methods to examine until now, and among that, microarray chip is noticed because of the various advantages. In this field, the development of molecularly organized sensing surface, on which high density of functionally active ligand groups with high sensitivity, selectivity, and orientation are arrayed, is a most important point. So, on the epoxy-activated glass, the widely usable chip platform was constructed with PAMAM dendrimer and strong neutravidin-biotin interaction, Also, to observe the usability of the constructed chip platform, protein microarray tests were executed.
