Hepatitis B virus(HBV) has been recognized to be the cause of hepatitis B and numerous studies about its pathogenesis and viral characteristics have been accomplished. However, currently there is no appropriate in vitro system for the study of HBV infection. Only some cell lines, such as HepG2, can be used for the viral replication when transiently or stably transfected with HBV DNA. So, in contrast to well-documented assembly and export of newly synthesized viral particles from the host cell, almost nothing is known about the early infection mechanism. Only the hepatocyte attachment site on the viral surface protein was identified as preS1(21-47) region. However, not yet the receptor for HBV has been isolated. Since the Hepatitis B viron is known to be internalized by receptor-mediated endocytosis via their envelope proteins, it is thinkable that these proteins also play a role in the intracellular transport of the virus. So, to evaluate whether the surface proteins, especially preS region, is involved in the early infection process containing initial attachment and intracellular transport of HBV genome into the host nucleus, the functions of preS region were analyzed. First, the interaction between preS region and polymerized human serum albumin (pHSA) was studied by competitive binding inhibition assay and the structural features of preS2 region contributed to the binding of pHSA was analyzed. And its importance in viral infection and vaccination would be discussed. Second, to investigate another role of preS region in the infection mechanism, intracellular distribution of exogenous pre S protein was visualized by confocal microscopy. The preS proteins were specifically accumulated into the nucleus and this process is energy- and mediator independent.
