Separation of particles has many application fields in the bio-chemical research such as purification of sample for chemical analysis, blood cells or blood plasma separation, separation of infected cell s from normal cells, DNA sorting and so on. The most bio-compatible separating method is radiation force driven separation due to its non-contact feature. However, previous methods of optical separation of particles are limited by their low throughput or non-continuous trait. In present study, a novel optical particle separating method is investigated theoretically and experimentally, to improve the previous optical separating methods. For the bio-chemical application, transparent particles having size range of 1 ~ 100 μm are considered. In the fundamental study, the analytic expressions for the radiation force on a sphere in a loosely focused Gaussian beam are derived in suitable form for applications by the photon stream method in ray optics regime. For perfect reflecting spheres, an analytic solution for the scattering force and a simplified expression for gradient force are obtained. And, an in situ separation system, cross-type optical chromatography, is developed theoretically and an analytic solution of the retention distance is derived. Particle trajectories in the cross-type optical chromatography are calculated for various sizes and materials of the particles, and flow velocities. Finally, an optical differential mobility analyzer for separating colloidal particles using a loosely focused beam is proposed theoretically and the concept of optical mobility is introduced. For the proposed optical differential mobility analyzer, particle trajectories and retention behavior are discussed and a transfer function described by operational parameters is derived.
