Rapid and accurate diagnosis of clinically pathogenic bacteria is important for the administration of proper antibiotics. This study reported development of microarray-based assay system for the high throughput identification of 44 clinically important pathogenic bacteria. Capture probes have designed for the detection of bacteria in general as well as specific target bacterial species. This oligonucleotide sequences were designed by bioinformatics analysis on the DNA sequences of 23S ribosomal DNA and 16S-23S spacer region. They were microarrayed on the slide glass and hybridized with target DNA isolated and amplified from a variety of clinical samples including reference bacteria. This microarray system was confirmed with hybridization results determining specificity and sensitivity of species-specific probe tested. Probes in this study had showed 100% specificity and positive predictive value. The microarray-based assay comprising DNA fragments detectable only specific bacteria is enable to diagnose pathogens more accurately, rapidly, easily, and cheaply compared with traditional method.
