Effective diagnosis of Korean Wilson disease by using oligonucleotide microarray system한국인 윌슨씨 병의 효과적인 진단을 위한 DNA 칩 시스템의 개발

I have developed a method for fabricating DNA microarrays that use a codon scanning algorithm to design oligonucleotide probes and pin spotting to immobilize 3’-terminal-aminated oligonucleotides on a glass surface. The oligonucleotides are covalently attached to the glass surface by a 3’ amino-linker that reacts with the aldehyde group on the glass surface. Using this method, I fabricated DNA chips that can detect 16 mutations containing all reported Korean substitution mutations in Wilson disease. These mutations were successfully detected with fluorescently labeled synthetic oligonucleotide reporters. Furthermore, three normal persons and nine Wilson disease patients were successfully diagnosed by hybridization of the PCR products obtained from the selected regions of genomic DNA. This research provides a simple, economical, rapid and efficient method for DNA chip production using the synthesized oligonucleotide.
