Recruited immune cells within the tumor microenvironment, have become immunosuppressive. Especially, tumor associated macrophages, TAMs are the most abundant cell type within the tumor site and are involved in various mechanisms of tumor biology, which include oncogenesis, drug resistance, as well as tumor metastasis. Thus, many researchers have studied on methods to reprogram macrophages with various activators. In general, macrophages are activated by toll-like receptor ligands. However, these molecules can induce side effects in body systemically, when they are injected directly. To reprogram the local macrophages without side effect, controllable supply of the damage-associated molecular patterns, DAMP molecules may solve the problem. Therefore, Indocyanine green-loaded red blood cell was used in this paper, to achieve that purpose. It can be disrupted under certain laser exposure and release the molecules. Through this, molecules can be delivered in a controlled manner to the restricted tumor site, reprogramming macrophages. A simple combination of bio-compatible molecules can enhance the function of the innate immune system locally and prevent tumor recurrence and metastasis.
