Identification of homologous recombination repair deficiency through the transcript usage analysis in breast cancer patients유방암 환자에서 전사체 분석을 통한 상동재조합수리 결핍의 확인

In Breast Cancer patients, Homologous Recombination deficiency(HRD) is an important factor for prediction drug response and prognosis. Many researchers suggested that somatic alteration or expression pattern of BRCA1/2 and RAD51C is biomarkers for HRD. However, the prediction of drug response and prognosis in patients without these events have been limited. In this study, I classified to HRD and Non-HRD group in 394 BRCA active patients. These groups showed significantly different splicing pattern, these genes were enriched DAN repair and response to DNA damage pathway. To shed light on the reason why BRCA active samples have high HRD, conducted an transcript usage(TU) analysis. Interestingly, significantly different TU was found between HRD and Non-HRD group. Finally, confirmed that these TU were highly correlated with drug response. Sum of that indicate that TU can be used to new biomarker for HRD in BRCA active samples.
