The study of the enzymes for production of muconic acid in corynebacterium코리네박테리움에서 뮤콘산 생합성을 위한 효소에 관한 연구

Adipic acid is precursor for the production of nylon and plastics. Today it is synthesized by chemical processes from petrochemicals. Biosynthesis of cis,cis-muconic aicd from chrosimate using aromatic degradation and assimilation in Corynebacterium glutamicum will replace the chemical synthetic method. Muconic acid biosynthesis pathway pass through three stages, protocatechuic acid and catechol into cis,cis-muconic acid. it can be chemically hydrogenated to adipic acid. The pathway consists of three enzymes, PobA(p-hydroxybenzoic acid hydrolase), AroY(protocatechuic acid decarboxylase) and CatA(catechol 1,2-dioxygenase). For each reaction step, we searched enzymes showing strong activity and collected several candidated enzymes of specific microorganisms. Then catalytic activity of the enzymes were tested and determined whether they can be introduced to the system of muconic acid production. To maximize productivity of muconic acid, the combinations of enzymes applied to various promoters were inserted pCES208 vector and the expression level were tested in C. glutamicum. This data will be applied to maximize production of muconic acid in C. glutamicum. This study can be introduced to designing and development of microbial systems which produce biomaterials aromatic compounds with high efficiency.
