Membrane cell recycle has been a very popular method in achieving a high bioreactor productivity. However, this process has several drawbacks such as difficulties of scale-up, sterilization, and fouling. In order to overcome these drawbacks, high cell density culture study of microorganisms was carried out using a membrane-less cell recycle fermenter. A new internal packing system which allowed microbial separation inside the fermenter was introduced and its applications for hight cell density culture of yeast and high ethanol productivity were investigated. The packed tube performance is very important in this system and is highly influenced by porosity of packing materials. The porosity of packing materials, 30~35 %, was suitable for yeast separation. Ethanol fermentation of Saccharomyces cerevisiae ATCC # 24858 was carried out and maximum cell concentration up to 87 g/L was obtained. The cell concentration in the fermentor was maintained by proper manipulation of dilution rate and natural bleed ratio through the packed tube. About 10 ~ 15 % of cells passed through the packed tube filter. This has the same concept as bleed ratio 0.1 ~ 0.15. The ethanol productivity with a feeding glucose concentration of 100 g/L was 21 g/Lㆍh at steady state. The internal packing system was successfully operated for more than 8 days. In the simulation of continuous ethanol fermentation in the packing system, it was possible to predict the glucose, ethanol, and yeast concentration. The predict values were in good agreement with experimental results. These studies show that the internal packing system can be used successfully for high cell density culture and ethanol fermentation, which was comparable with an external membrane cell recycle system.
